Isolasi Bakteri Bacillus thuringiensis dari Tanah Kota Makassar dan Uji Aktivitas Bioinsektisida terhadap Larva Nyamuk Aedes aegypti


An investigation of bacteria
Bacillus thuringiensis
from soil in Makassar have
been done. This study aims to obtain bacterial isol
ates of
Bacillus thuringiensis
as a
potential biological control against diseases cause
d by vector Aedes aegypti.
Bacillus thuringiensis
bacteria isolated from soil by using Ohba and Aiza
wa method.
The medium used are Nutrient Agar (NA) medium and L
uria Bertani 0.25 M acetate
buffer pH 6.8 as selective media. Isolates were the
n characterized and identified based
on colony and cell morphology, Gram staining, spore
staining, and biochemical tests,
these isolates were identified as members of
Bacillus thuringiensis
. Total of 19 colonies
from 3 soil samples were analyzed and 1 colony of
Bacillus thuringiensis
obtained,
which colonies B

9 were showing positive results as
Bacillus thuringiensis.
Further testing biopestiside activity using third i
nstar larvae of
Aedes aegypti
were
divided into 6 groups, namely the 5 treatment group
s were given fermented by mixing
the sediment into the water to obtain a concentrati
on of 5%, 10%, 15%, 20%, 25% and 1
control group in the form of distilled water.
The test results showed that these isolates can cau
se mortality ranged from 6,67- 60%
after observation for 24 hours and the highest mort
ality occurred at a concentration of
25% with LC
50
value was 24,62%.
